MOTION randomised 123 patients with symptomatic tenosynovial giant cell tumour not suitable for surgery to the CSF1R inhibitor vimseltinib or placebo. Tumours shrank in 40% on vimseltinib versus none on placebo, with better joint movement, pain and function, leading to approval in February 2025; whether responses last after stopping is unknown.
MOTION, trial NCT05059262 sponsored by Deciphera and published in the Lancet in 2024, showed that the CSF1R inhibitor vimseltinib shrank tenosynovial giant cell tumours in forty percent of patients versus none on placebo, with better joint function. It randomised 123 patients with symptomatic disease not amenable to surgery, met its primary objective response endpoint at week 25 and improved active range of motion, pain, stiffness and function, leading to approval in February 2025. OnCo links it to sarcomas and tenosynovial giant cell tumour and to vimseltinib. Whether long-term treatment is needed or responses persist after stopping is the open question for a benign but disabling tumour.
