Ripretinib kept fourth-line GIST under control six times longer than placebo and improved survival.
INVICTUS, trial NCT03353753 sponsored by Deciphera and published in Lancet Oncology in 2020, showed that the switch-control kinase inhibitor ripretinib keeps fourth-line gastrointestinal stromal tumour under control roughly six times longer than placebo and improves survival. It randomised 129 patients after at least three kinase inhibitors and met its primary progression-free survival endpoint with a very large effect and a survival benefit, leading to approval in May 2020. OnCo links it to sarcomas and GIST, KIT as a target and ripretinib. Whether ripretinib can move earlier in the sequence, which INTRIGUE and the mutation-selected INSIGHT trial address, is the open question.
