Nouscom is developing Nous-209, an off-the-shelf viral-vector vaccine encoding 209 shared frameshift neoantigens to intercept cancer in Lynch syndrome carriers.
Nouscom, based in Basel and Rome, is developing Nous-209, an off-the-shelf viral-vector vaccine encoding 209 shared frameshift neoantigens, to intercept cancer in people with Lynch syndrome. A phase 1b/2 in Lynch carriers, published in Nature Medicine in March 2026, showed safety and immune responses in most participants, re-treatment and long-term follow-up data were selected for an ESMO 2026 oral, and the vaccine has also been tested with pembrolizumab in microsatellite-instability-high tumours. OnCo links it to colorectal cancer, to off-the-shelf cancer vaccines and chemoprevention, to the MSI-H and neoantigen terms, and to the idea of a randomised trial of a shared-antigen vaccine to prevent Lynch syndrome cancers. Whether immune responses translate into fewer cancers in carriers is the question only a prevention trial can answer. The off-the-shelf vaccine page carries the field.
