Built the pancreatic cancer organoid models now used to test drugs against a patient's own tumour.
David A. Tuveson is Director of the Cold Spring Harbor Laboratory Cancer Center and Roy J. Zuckerberg Professor, specialising in pancreatic ductal adenocarcinoma. He is known for building the first pancreatic ductal organoid models, now used to test drugs against a patient's own tumour, and the mouse models that defined stromal biology in the disease. His selected papers describe organoid models of human and mouse ductal pancreatic cancer and organoid profiling that identifies common responders to chemotherapy. His laboratory drives organoid pharmacotyping trials and functional ex vivo drug testing, and he is a Lustgarten Foundation leader.

It explains why single-sample classifiers disagree on about one tumour in eight and why KRAS allelic imbalance and GATA6 copy number are being read alongside expression.
Fibroblasts are part of the immune conversation, not just a physical barrier, which is why stromal and immune strategies are now designed together.
Together with COMPASS it made sequencing at diagnosis of advanced disease a standard expectation, and it found the BRAF-deletion class that a hotspot test misses.
The myCAF and iCAF distinction is why 'target the stroma' became 'target a fibroblast state', and IL-6 blockade combinations follow from it.
The mechanistic account of T-cell exclusion that all later combination immunotherapy trials in pancreatic cancer cite, and the origin of CXCR4 inhibitor combinations.
It launched a decade of attempts to strip the stroma to let drugs in; the clinical trials that followed failed, and later mouse work showed the stroma also restrains the tumour.
