Andrew J. Aguirre is an Institute Member at the Broad Institute of MIT and Harvard and a physician at Dana-Farber, working on pancreatic ductal adenocarcinoma and KRAS biology. He is known for leading real-time genomic profiling of advanced pancreatic cancer to enable precision medicine, the subject of his selected paper. His laboratory uses patient-derived organoids and functional genomics to find what pancreatic cancers depend on beyond KRAS. He now studies RAS-pathway dependencies and resistance to KRAS and RAS inhibitors.

A germline result is not the whole story: without loss of the second allele the tumour may not be repair-deficient, which is why tumour sequencing and, in trials, HRD signatures are read alongside.
Together with COMPASS it made sequencing at diagnosis of advanced disease a standard expectation, and it found the BRAF-deletion class that a hotspot test misses.
It is the reference multi-platform dataset and the reason a KRAS wild-type report is treated as a search for another driver rather than as an absence.
