PROMID, trial NCT00171873 sponsored by Novartis and the German NET study group and reported in 2009, showed for the first time that a hormone-suppressing somatostatin analogue injection also slows neuroendocrine tumour growth. It randomised 85 patients with treatment-naive metastatic midgut neuroendocrine tumours to octreotide LAR or placebo and met its primary endpoint of time to tumour progression with a large effect, while overall survival did not differ because of crossover and a low event rate. OnCo links it to neuroendocrine tumours and to the somatostatin analogues record, and CLARINET extended the antiproliferative effect to lanreotide across enteropancreatic tumours. Whether starting a somatostatin analogue early changes survival, rather than only delaying progression, remains unproven.
