DREAMM-8 is the second confirmatory win for belantamab, in lenalidomide-exposed patients.
DREAMM-8, trial NCT04484623 sponsored by GSK and published in the New England Journal of Medicine in 2024, is the second confirmatory win for belantamab mafodotin, in relapsed myeloma patients after at least one line including lenalidomide. It randomised 302 patients to belantamab with pomalidomide and dexamethasone or bortezomib with the same partners and met its primary progression-free survival endpoint at twelve months with a favourable survival trend, supporting the 2025 US approval with pomalidomide and dexamethasone. OnCo links it to multiple myeloma, belantamab mafodotin and Sagar Lonial, and the result is concordant with DREAMM-7. Whether the pomalidomide combination or the bortezomib combination is the better use of the drug is the open question.
