In AETHERA, a year of the CD30 ADC after transplant halved relapse risk in high-risk patients.
AETHERA, trial NCT01100502 sponsored by Seagen and published in the Lancet in 2015, showed that a year of the CD30 antibody-drug conjugate brentuximab vedotin after autologous transplant halved the risk of relapse in high-risk Hodgkin lymphoma. It randomised 329 patients to brentuximab consolidation or placebo and met its primary progression-free survival endpoint with a large effect, while overall survival did not differ because of crossover, leading to approval in August 2015 as post-transplant consolidation. OnCo links it to Hodgkin lymphoma, autologous transplant and brentuximab vedotin. Now that brentuximab and PD-1 blockade are used before transplant, whether consolidation still adds anything is the open question.
