A single change in a histone protein that defines diffuse midline glioma, the childhood brain tumour with the fewest treatment options, and now the target of the first approved drug for it.
Lysine-to-methionine mutation at position 27 of histone H3 (H3F3A or HIST1H3B) causes global loss of H3K27 trimethylation. Defines WHO grade 4 diffuse midline glioma (including DIPG), median survival ~11 months, radiotherapy the only proven therapy until dordaviprone (2025). GD2 is overexpressed, enabling CAR-T.
Showing the molecule this term concerns: Dordaviprone.
The glossary entry explains the word; the readout page carries the scoring rule, the thresholds approvals use, the companion diagnostics and the tests.
