The anti-CCR4 antibody mogamulizumab doubled the time to progression compared with vorinostat in previously treated mycosis fungoides and Sezary syndrome, with particularly strong activity in the blood, and became the first antibody approved for these lymphomas.
Phase 3 trial of 372 patients with relapsed or refractory mycosis fungoides or Sezary syndrome after at least one systemic therapy randomised to mogamulizumab or vorinostat.
Median progression-free survival was 7.7 versus 3.1 months (hazard ratio 0.53), global response 28 versus 5 percent, with blood responses in 68 percent of mogamulizumab patients; drug rash was the main toxicity.
Mogamulizumab is a standard for advanced-stage cutaneous T-cell lymphoma with blood involvement, particularly Sezary syndrome.
