Tracon was a San Diego biotech whose endoglin antibody carotuximab failed a phase 3 trial in angiosarcoma and whose subcutaneous PD-L1 antibody envafolimab missed in sarcoma; it wound down in 2024.
Tracon Pharmaceuticals, based in San Diego, developed carotuximab (TRC105), an antibody against endoglin on tumour blood vessels, which failed the phase 3 TAPPAS trial with pazopanib in angiosarcoma in 2019, and then in-licensed the subcutaneous PD-L1 antibody envafolimab, whose ENVASARC trial in undifferentiated pleomorphic sarcoma did not meet its endpoint in 2023. The company ceased operations in 2024. Its trials remain on ClinicalTrials.gov under Tracon as lead sponsor.
Envafolimab is the first PD-L1 antibody given as a quick injection under the skin rather than an infusion, approved in China for advanced tumours with mismatch-repair deficiency.
Carotuximab was an antibody against endoglin, a protein on growing blood vessels and on angiosarcoma cells. Added to pazopanib in the phase 3 TAPPAS trial it did not help, and the trial was stopped in 2019.
