PROTAC pioneer whose vepdegestrant became the first approved targeted protein degrader in 2026.
Arvinas, based in New Haven and listed as ARVN, is the PROTAC pioneer whose vepdegestrant became the first approved targeted protein degrader in 2026. Founded on Craig Crews' PROTAC technology, it developed vepdegestrant, an oestrogen receptor degrader, with Pfizer, and its pipeline includes ARV-393 against BCL6 and ARV-102 against LRRK2 in neurology. OnCo links it to HR-positive, HER2-negative breast cancer, to PROTACs and molecular glues, to the bottleneck of undruggable drivers, and to ideas for an open degrader consortium and for destroying the truncated androgen receptor. Whether degraders can reach transcription factors that inhibitors never could is the open question the first approval makes urgent. Vepdegestrant has its own page.
Vepdegestrant is the first PROTAC ever approved (2026): a pill that tags the oestrogen receptor for destruction, for breast cancers with ESR1 mutations.
