Arsenal Biosciences, based in South San Francisco, is a programmable CAR-T company that uses logic gates so its cells fire only when two tumour proteins are present, and its AB-2100 is in phase 1/2 for kidney cancer. AB-2100 combines a synthetic AND gate requiring both PSMA and CA9, shRNA armouring against suppression in the tumour microenvironment and a synthetic pathway activator, and is being tested in clear-cell renal cell carcinoma after immunotherapy and VEGF therapy in trial NCT06245915; AB-1015 targets ovarian cancer, and the company raised 325 million dollars with Regeneron and Bristol Myers Squibb participating. OnCo links it to renal and ovarian cancer, to armoured and logic-gated CARs and logic-gated therapeutics, and to PSMA as a target. Whether logic gating solves the on-target, off-tumour problem that has stalled solid-tumour CAR-T is the question its trial will test.
