HERA, NSABP B-31 and NCCTG N9831, led by trial NCT00045032 and sponsored by BIG, NSABP and NCCTG, were three adjuvant trials reported together in 2005 that showed a year of trastuzumab after surgery cuts the risk of dying from HER2-positive breast cancer by about a third. Across roughly 9,000 women the joint B-31 and N9831 analysis and the HERA trial both showed large, durable improvements in disease-free and overall survival at ten and eleven years despite heavy crossover in HERA, turning the worst breast cancer subtype into one of the most curable, and HERA also showed that two years of trastuzumab is no better than one.
