ATIC (Bifunctional purine biosynthesis protein ATIC) is an enzyme. In the public catalogues the evidence so far is association rather than a proven role. Tied to Lung cancer.
Bifunctional enzyme that catalyses the last two steps of purine biosynthesis. Acts as a transformylase that incorporates a formyl group to the AMP analog AICAR (5-amino-1-(5-phospho-beta-D-ribosyl)imidazole-4-carboxamide) to produce the intermediate formyl-AICAR (FAICAR). Can use both 10-formyldihydrofolate and 10-formyltetrahydrofolate as the formyl donor in this reaction.
Open Targets scores its association with cancer at 0.71 (direct and indirect evidence; datatypes literature 0.93, affected pathway 0.93, genetic association 0.00, somatic mutation 0.90).

No product in this corpus aims at ATIC yet. Inhibitors are shaped to fit the enzyme's active site so the reaction the cancer relies on stops.
First described 1992. Earliest sequence paper UniProt cites for the protein: Rasmussen H.H. et al, Electrophoresis, 1992, "Microsequences of 145 proteins recorded in the two-dimensional gel protein database of normal human epidermal keratinocytes". Source.

Can use both 10-formyldihydrofolate and 10-formyltetrahydrofolate as the formyl donor in this reaction. Also catalyses the cyclisation of FAICAR to inosine monophosphate (IMP). Is able to convert thio-AICAR to 6-mercaptopurine ribonucleotide, an inhibitor of purine biosynthesis used in the treatment of human leukaemias. Promotes insulin receptor/INSR autophosphorylation and is involved in INSR internalisation. Location: Cytoplasm, cytosol (UniProt). Locus 2q35 (HGNC).
