Overview
Zymeworks, based in Vancouver and listed as ZYME, invented zanidatamab, the HER2 bispecific antibody, and a hydrophilic topoisomerase 1 ADC platform. Its Azymetric platform produced zanidatamab, on which it earns royalties, and its own pipeline includes ZW191, a folate receptor alpha ADC that reported a 52 percent response rate in pretreated ovarian and endometrial cancer at AACR 2026, and ZW251 against GPC3. OnCo links it to biliary tract cancer and HER2-positive breast cancer, to the zanidatamab drug record and to Zai Lab. Whether its second-generation ADCs can repeat the success of a bispecific it no longer controls is the open question. Zanidatamab has its own page.

Zanidatamab (Ziihera) is an antibody that grabs HER2 at two different spots, approved for HER2+ bile duct cancer.
Phase 1Biparatopic HER2 antibody-drug conjugate
Zanidatamab zovodotin joined the two-armed HER2 antibody zanidatamab to a cell-killing payload. It was tested in early trials in HER2-expressing cancers; the plain antibody went on to approval, the conjugate did not progress beyond phase 1.
