RAS/MAPK-pathway company whose avutometinib + defactinib combination became the first approved treatment for KRAS-mutant low-grade serous ovarian cancer (May 2025).
Verastem Oncology, based in Needham, Massachusetts, and listed as VSTM, is a RAS and MAPK pathway company whose combination of avutometinib and defactinib became the first approved treatment for KRAS-mutant low-grade serous ovarian cancer in May 2025. The AVMAPKI FAKZYNJA CO-PACK, pairing the RAF and MEK clamp avutometinib with the FAK inhibitor defactinib, received accelerated approval on 8 May 2025 on the RAMP 201 trial, and the company is also developing a KRAS G12D inhibitor, VS-7375, and combinations in pancreatic cancer with GenFleet. OnCo links it to ovarian and pancreatic cancer, to kinase and KRAS inhibitors, to KRAS as a target and the MAPK pathway, and to the RAMP 201 trial and the combination drug record. Whether the combination extends into pancreatic cancer is the open question. Avutometinib plus defactinib has its own page.
VS-7375 is an experimental small-molecule drug from Verastem in phase 2 trials for pancreatic ductal adenocarcinoma, non-small-cell lung cancer and colorectal cancer, aimed at KRAS.
Avutometinib plus defactinib is the first treatment approved specifically for low-grade serous ovarian cancer, a slow-growing type driven by the RAS pathway.
