Nested Therapeutics is a Cambridge, Massachusetts precision oncology company founded by Versant Ventures. Its platform maps mutational clusters onto protein structures to reveal cryptic driver mutations and druggable pockets, combining genomics, computational biophysics and machine learning. The lead programme, NST-628, is a non-degrading, brain-penetrant pan-RAF/MEK molecular glue that outperformed single agents and vertical combinations in RAS/MAPK-driven preclinical models; it is in a phase 1 study (NCT06326411) in adults with MAPK pathway-mutated or -dependent advanced solid tumours. A mutation-selective p53 pathway programme targets a cryptic pocket in a transcriptional regulator and has a lead compound. Nested launched in October 2022 with a 90 million US dollar Series A led by Goldman Sachs Asset Management, bringing total funding to 125 million US dollars.
