Glembatumumab vedotin (CDX-011, GPNMB-directed MMAE ADC) did not meet its primary endpoint in the METRIC trial in GPNMB-overexpressing metastatic TNBC (2018). Celldex's lead asset is now barzolvolimab, an anti-KIT antibody in phase 3 for chronic urticaria; its oncology footprint is historical but instructive for ADC target selection.
Rindopepimut is a peptide vaccine against EGFRvIII, a mutant protein found only on some glioblastomas. After a phase 2 that beat historical controls, the 745-patient double-blind phase 3 ACT IV found no benefit in 2016, and tumours in both arms had lost EGFRvIII at recurrence, a lesson in antigen escape.
