Blocking cortisol signalling in tumour cells made chemotherapy work better and extended life in resistant ovarian cancer.
ROSELLA, also GOG-3073, trial NCT05257408 sponsored by Corcept and published in the Lancet in 2025, showed that blocking cortisol signalling in tumour cells with relacorilant makes nab-paclitaxel chemotherapy work better and extends life in platinum-resistant ovarian cancer. It randomised 381 women with one to three prior lines to relacorilant plus nab-paclitaxel or nab-paclitaxel alone, met its primary progression-free survival endpoint and improved overall survival, leading to FDA approval in the first quarter of 2026 as Lifyorli, the first-in-class glucocorticoid receptor antagonist in oncology. OnCo links it to ovarian cancer, relacorilant, paclitaxel and Corcept Therapeutics, and the record notes it as a chemosensitisation mechanism rather than a new cytotoxic. Whether the approach transfers to other chemotherapy-treated cancers is the open question.
