Tests whether a CDH6-targeting ADC with Enhertu's payload can beat chemotherapy in resistant ovarian cancer.
REJOICE-Ovarian01, trial NCT06161025 sponsored by Daiichi Sankyo and Merck, is the phase 2/3 test of whether raludotatug deruxtecan, a CDH6-targeting antibody-drug conjugate carrying the same payload as trastuzumab deruxtecan, can beat physician's choice chemotherapy in platinum-resistant ovarian cancer after one to three prior lines. The phase 2 dose-optimisation part reported an objective response rate of 50.5 percent at ESMO 2025, the phase 3 part randomises at 5.6 mg/kg against chemotherapy, and readout is expected in 2026 to 2027. OnCo links it to ovarian cancer, antibody-drug conjugates, CDH6 as a target and raludotatug deruxtecan. Whether a new ADC target can match mirvetuximab's survival benefit, and in which patients, is the question it will answer.
