The first ADC to extend life in ovarian cancer, for the roughly one-third of tumours with high folate receptor alpha.
MIRASOL, also GOG-3045, trial NCT04209855 sponsored by ImmunoGen, now AbbVie, and published in the New England Journal of Medicine in 2023, produced the first antibody-drug conjugate to extend life in ovarian cancer, for the roughly one-third of platinum-resistant tumours with high folate receptor alpha expression. It randomised 453 women with one to three prior lines to mirvetuximab soravtansine or chemotherapy, met its primary progression-free survival endpoint and improved overall survival, leading to full approval in March 2024; ocular toxicity is frequent but reversible with eye-drop prophylaxis. OnCo links it to ovarian cancer, antibody-drug conjugates, folate receptor alpha, mirvetuximab soravtansine, Kathleen N. Moore, Ursula A. Matulonis and the GOG Foundation. Whether the drug can move into platinum-sensitive or maintenance settings is the open question.
