MEVPRO-1 is the phase 3 trial of the first EZH2 inhibitor combination in prostate cancer.
MEVPRO-1, trial NCT06551324 sponsored by Pfizer, is the phase 3 trial of the first EZH2 inhibitor combination in prostate cancer, testing mevrometostat plus enzalutamide against physician's choice of enzalutamide or docetaxel in metastatic castration-resistant prostate cancer after abiraterone. It enrols about 600 men in an open-label design with radiographic progression-free survival as the primary endpoint; the first patient was dosed in October 2024 and completion is projected for 2028, while MEVPRO-2, a double-blind trial, and MEVPRO-3 in castration-sensitive disease run in parallel. OnCo links it to EZH2 as a target and to mevrometostat and enzalutamide. There are no results, and whether epigenetic reversal of androgen receptor resistance works in patients is the question the programme exists to answer. Mevrometostat has its own page.
