MagnetisMM-3, trial NCT04649359 sponsored by Pfizer and reported in 2023, is the pivotal single-arm study behind elranatamab's approval in relapsed or refractory myeloma after at least three prior lines in BCMA-naive patients. In 123 patients the response rate was 61 percent with complete response or better in 35 percent, progression-free survival was durable, and switching to every-two-week dosing after six months maintained responses. OnCo links it to multiple myeloma, elranatamab and the MagnetisMM-3 paper. Whether reduced dosing frequency lowers infection risk without losing efficacy is the open question its design raised.
