The first head-to-head trial among KRAS drugs: Roche's divarasib beat the two approved pills on progression and survival.
Krascendo 1, trial NCT06497556 sponsored by Roche, is the first head-to-head trial among KRAS drugs, comparing divarasib with sotorasib or adagrasib in previously treated KRAS G12C-mutant non-small-cell lung cancer. Roche announced on 2 July 2026 that divarasib showed superior progression-free survival by blinded review and superior overall survival at an interim analysis against the two approved inhibitors, with full data expected at a 2026 congress from a trial of 338 patients. OnCo links it to KRAS as a target and to all three drugs, and notes that Krascendo 2 tests divarasib with pembrolizumab in the first line. Because the numbers are pending and this is the first such comparison, there is no independent replication, and whether a more potent G12C inhibitor changes the modest picture left by CodeBreaK 200 and KRYSTAL-12 is the open question.
