IZABRIGHT-Breast01, trial NCT06926868 sponsored by Bristol Myers Squibb, is the global first-line trial of the EGFR by HER3 bispecific antibody-drug conjugate izalontamab brengitecan in metastatic triple-negative breast cancer patients ineligible for PD-1 or PD-L1 inhibitors, comparing it with chemotherapy. It plans to enrol 570 patients, directly competes for the populations studied in ASCENT-03 and TROPION-Breast02, and is ongoing with no results. OnCo links it to triple-negative breast cancer, izalontamab brengitecan and the TNBC roadmap. Whether a bispecific ADC that succeeded in a Chinese phase 3 can match or beat the approved TROP2 ADCs in a global first-line population is the question it exists to answer.
