CARTITUDE-5, trial NCT04923893 sponsored by Janssen, tests ciltacabtagene autoleucel as frontline consolidation instead of ongoing lenalidomide maintenance in newly diagnosed myeloma patients for whom transplant is not planned, after bortezomib, lenalidomide and dexamethasone induction. Together with CARTITUDE-6, known as EMagine, which compares cilta-cel with autologous transplant after daratumumab-based induction in transplant-eligible patients, it asks whether one CAR-T infusion can replace transplant or maintenance, and there are no results yet. OnCo links it to multiple myeloma, autologous transplant, cilta-cel, the idea of one CAR-T infusion instead of transplant and the CARTITUDE-4 paper. Whether a single infusion can match years of maintenance is the question it exists to answer.
