A third PARP inhibitor showed the same pattern: longer disease control after first-line chemotherapy, biggest in tumours with faulty DNA repair.
ATHENA-MONO, also GOG-3020, trial NCT03522246 sponsored by Clovis, now pharma&, and published in JCO in 2022, showed that a third PARP inhibitor follows the same pattern as the others: rucaparib maintenance after response to first-line platinum chemotherapy in newly diagnosed advanced ovarian cancer prolongs disease control, most in tumours with faulty DNA repair. It randomised 538 patients to rucaparib or placebo and met its primary progression-free survival endpoint in the whole population with a larger effect in homologous recombination-deficient disease. OnCo links it to ovarian cancer, PARP as a target, rucaparib and Bradley J. Monk. Clovis went bankrupt in 2022 and US first-line maintenance labelling did not follow, so whether rucaparib ever takes a place beside olaparib and niraparib in first-line maintenance is the open question.
