SpringWorks Therapeutics, based in Stamford, Connecticut, developed nirogacestat, the first approved treatment for desmoid tumours, and mirdametinib for NF1 plexiform neurofibromas, and Merck KGaA completed its acquisition of the company for around 3.9 billion dollars in 2025. Nirogacestat, sold as Ogsiveo, was approved in November 2023 on the DeFi trial, and mirdametinib, sold as Gomekli, in February 2025. OnCo links it to desmoid tumour and sarcomas, to both drug records, and to the bottleneck of rare and paediatric cancers without markets, which its two products show can be overcome. Whether a large pharma keeps investing in rare-tumour indications of this size is the open question after the acquisition. Nirogacestat and mirdametinib have their own pages.

Nirogacestat is the first approved medicine for desmoid tumours, locally invasive growths that do not spread but can be crippling; it works by blocking Notch signalling.
Mirdametinib (Gomekli in the US, Ezmekly in Europe) is a MEK-blocking capsule or dispersible tablet for adults and children with neurofibromatosis type 1 whose plexiform neurofibromas cannot be removed surgically; it is the first such drug approved for adults.
