Rhythm Pharmaceuticals is a Boston company whose MC4R agonist setmelanotide, sold as Imcivree, treats the uncontrolled hunger and obesity of rare genetic disorders and of damage to the hypothalamus, including after treatment for craniopharyngioma and other brain tumours.
Rhythm Pharmaceuticals develops treatments for rare diseases of the melanocortin-4 receptor pathway that controls hunger. Setmelanotide (Imcivree, RM-493) is approved in the United States and Europe for obesity due to POMC, PCSK1 or LEPR deficiency and for Bardet-Biedl syndrome. Its phase 3 TRANSCEND trial in acquired hypothalamic obesity, which follows surgery or radiotherapy for craniopharyngioma and other suprasellar tumours, met its primary endpoint in 2025 and supports a supplemental filing.
Setmelanotide is a daily injection that switches back on the brain's fullness signal. It is approved for rare genetic causes of obesity and, since March 2026 in the United States, for the continuous weight gain that follows damage to the hypothalamus from a tumour such as craniopharyngioma or from its treatment.
