Nektar is a San Francisco biotechnology company whose polymer conjugation chemistry underlies several approved drugs and whose engineered interleukin-2, bempegaldesleukin, failed in phase 3 with nivolumab in 2022.
Nektar Therapeutics, listed on Nasdaq, built its business on PEGylation and polymer conjugate chemistry licensed into products such as pegfilgrastim-type molecules and movantik. Its own oncology bet, bempegaldesleukin (NKTR-214), a PEGylated interleukin-2 designed to favour effector T cells, was developed with Bristol Myers Squibb in a large phase 3 programme with nivolumab that reported negative results in melanoma, renal cell carcinoma and bladder cancer in 2022, after which the programme was closed. OnCo records it as a registry sponsor of phase 3 cancer trials.
Bempegaldesleukin was a re-engineered interleukin-2 meant to be a safer version of a famous old immunotherapy. It added nothing to nivolumab in three phase 3 trials.
