It is more myelosuppressive and less neurotoxic than vincristine, which is why the two are chosen for different regimens. Approved in the US in 1965, it is the V in ABVD, the standard chemotherapy for Hodgkin lymphoma, and was in PVB, the cisplatin combination that first cured metastatic testicular cancer. It also features in MVAC for bladder cancer, in treatment of Kaposi sarcoma and Langerhans cell histiocytosis, and with carboplatin for low-grade glioma in children. It is a vesicant that causes severe tissue injury if it leaks from the vein, and myelosuppression is the dose-limiting toxicity.
