{"entity":{"id":"mezigdomide","kind":"drug","name":"Mezigdomide","aka":[],"tldr":"Mezigdomide is the most potent oral cereblon modulator, producing responses in about 40% of triple-class-refractory myeloma with dexamethasone alone.","summary":"Mezigdomide is the most potent oral cereblon E3 ligase modulator (CELMoD) in development, producing rapid and deep degradation of Ikaros and Aiolos with strong T-cell co-stimulation. In CC-92480-MM-001, mezigdomide with dexamethasone alone produced an ORR of 41% in heavily pretreated, triple-class-refractory myeloma, including responses in 30% of patients with extramedullary disease, a group that responds poorly to most therapies. Two phase 3 trials are ongoing: SUCCESSOR-1 compares Mezi-Vd with Pom-Vd, and SUCCESSOR-2 compares Mezi-Kd with Kd. The open questions are durability of response and whether cytopenias and infections limit its use in combination. BMS develops it alongside the related CELMoD iberdomide. It is a pill that acts on the same target as lenalidomide but strongly enough to work when that drug and its successors have failed.","status":"phase-3","asOf":"2026-09-07","links":[{"label":"ClinicalTrials.gov: trials of Mezigdomide","url":"https://clinicaltrials.gov/search?intr=CC-92480"}],"tags":[],"related":[],"cancers":["multiple-myeloma"],"sections":[],"technologies":["protac-degrader","celmods"],"targets":[],"drugs":[],"companies":["bms"],"institutions":[],"pathways":[],"terms":[],"trials":["nct05519085","nct05552976","nct05372354","nct03989414","nct06988488","nct06892522"],"people":[],"bottlenecks":[],"keyPapers":[],"journals":[],"dependsOn":[],"notes":[],"code":"CC-92480","modality":"CELMoD (cereblon E3 ligase modulator)","mechanism":"High-affinity cereblon modulator; rapid, deep Ikaros/Aiolos degradation with strong T-cell co-stimulation.","approvals":[],"mechanismSteps":[],"toxicity":[],"access":[],"regulatoryEvents":[]},"route":"/drugs/mezigdomide/","neighbours":{"cancer":[{"id":"multiple-myeloma","kind":"cancer","name":"Multiple myeloma","route":"/cancers/multiple-myeloma/"},{"id":"myeloma-relapsed-refractory","kind":"cancer","name":"Relapsed or refractory multiple myeloma","route":"/cancers/myeloma-relapsed-refractory/"}],"technology":[{"id":"celmods","kind":"technology","name":"Cereblon E3 ligase modulators (CELMoDs)","route":"/technologies/celmods/"},{"id":"protac-degrader","kind":"technology","name":"PROTACs & molecular glues (targeted protein degradation)","route":"/technologies/protac-degrader/"}],"company":[{"id":"bms","kind":"company","name":"Bristol Myers Squibb","route":"/companies/bms/"}],"trial":[{"id":"nct06892522","kind":"trial","name":"A Study to Assess Change in Disease Activity and Adverse Events (AE)s in Adult Participants With Multiple Myeloma Receiving T Cell Engaging Bi/Tri-spe","route":"/trials/nct06892522/"},{"id":"nct03989414","kind":"trial","name":"A Study to Determine the Recommended Dose and Regimen and to Evaluate the Safety and Preliminary Efficacy of CC-92480 in Combination With Standard Tre","route":"/trials/nct03989414/"},{"id":"nct06988488","kind":"trial","name":"A Study to Determine the Recommended Dose and Schedule, and Evaluate the Safety and Preliminary Efficacy of Mezigdomide in Combination With Elranatama","route":"/trials/nct06988488/"},{"id":"nct05552976","kind":"trial","name":"A Study to Evaluate Mezigdomide in Combination With Carfilzomib and Dexamethasone (MeziKD) Versus Carfilzomib and Dexamethasone (Kd) in Participants W","route":"/trials/nct05552976/"},{"id":"nct05519085","kind":"trial","name":"A Study to Evaluate Mezigdomide, Bortezomib and Dexamethasone (MEZIVd) Versus Pomalidomide, Bortezomib and Dexamethasone (PVd) in Participants With Re","route":"/trials/nct05519085/"},{"id":"nct05372354","kind":"trial","name":"A Study to Evaluate Safety, Drug Levels and Effectiveness of CC-92480 (BMS-986348) in Combination With Other Treatments in Participants With Relapsed ","route":"/trials/nct05372354/"}],"pathway":[{"id":"ubiquitin-proteasome-system","kind":"pathway","name":"Ubiquitin-proteasome system & protein homeostasis","route":"/pathways/ubiquitin-proteasome-system/"}]}}
