{"entity":{"id":"grid","kind":"trial","name":"GRID","aka":[],"tldr":"GRID showed that regorafenib held back GIST that had already escaped both imatinib and sunitinib, and gave patients a third line of targeted treatment.","summary":"GRID randomised 199 patients with metastatic or unresectable gastrointestinal stromal tumour that had progressed on imatinib and sunitinib two to one to regorafenib, a multikinase inhibitor of KIT, PDGFRA and VEGF receptors, or placebo, with crossover at progression. The primary endpoint was progression-free survival.\n\nRegorafenib lengthened progression-free survival several-fold compared with placebo, with disease control in most patients, at the cost of hand-foot skin reaction, hypertension and diarrhoea. Overall survival was confounded by crossover. Regorafenib was approved in the United States in February 2013 as third-line therapy for GIST and held that place until ripretinib arrived in 2020.","status":"positive","asOf":"2026-09-17","links":[{"label":"ClinicalTrials.gov NCT01271712","url":"https://clinicaltrials.gov/study/NCT01271712"}],"tags":["subtype-trials"],"related":[],"cancers":["gist-imatinib-resistant","gist","sarcoma"],"sections":[],"technologies":["antiangiogenic","kinase-inhibitors"],"targets":[],"drugs":["regorafenib"],"companies":["bayer"],"institutions":[],"pathways":[],"terms":[],"trials":[],"people":["george-demetri"],"bottlenecks":[],"keyPapers":["paper-grid-regorafenib-gist-lancet-2013"],"journals":[],"dependsOn":[],"notes":[],"nct":"NCT01271712","phase":"3","setting":"Metastatic or unresectable gastrointestinal stromal tumour after failure of imatinib and sunitinib: regorafenib against placebo","sponsor":"Bayer","result":"Regorafenib lengthened progression-free survival compared with placebo in GIST after imatinib and sunitinib; approved in February 2013.","started":"2011-01-04","startedType":"actual","yearReported":2013,"enrolled":199,"enrolledBasis":"registry","outcomes":[{"endpoint":"Progression-free survival (independent blinded central review)","primary":true,"unit":"months","arms":[{"name":"Regorafenib","n":133,"value":4.8},{"name":"Placebo","n":66,"value":0.9}],"hr":0.27,"ci":[0.19,0.39],"p":"<0.0001","source":"https://doi.org/10.1016/S0140-6736(12)61857-1"}]},"route":"/trials/grid/","neighbours":{"cancer":[{"id":"gist","kind":"cancer","name":"Gastrointestinal stromal tumour (GIST)","route":"/cancers/gist/"},{"id":"gist-imatinib-resistant","kind":"cancer","name":"Imatinib-resistant GIST","route":"/cancers/gist-imatinib-resistant/"},{"id":"sarcoma","kind":"cancer","name":"Sarcomas (soft tissue, bone, GIST)","route":"/cancers/sarcoma/"}],"technology":[{"id":"antiangiogenic","kind":"technology","name":"Anti-angiogenic therapy","route":"/technologies/antiangiogenic/"},{"id":"kinase-inhibitors","kind":"technology","name":"Small-molecule kinase inhibitors","route":"/technologies/kinase-inhibitors/"}],"drug":[{"id":"regorafenib","kind":"drug","name":"Regorafenib","route":"/drugs/regorafenib/"}],"company":[{"id":"bayer","kind":"company","name":"Bayer","route":"/companies/bayer/"}],"person":[{"id":"george-demetri","kind":"person","name":"George D.
