In phase 1/2, golcadomide with rituximab gave an ORR of about 50% in relapsed or refractory DLBCL, and with R-CHOP in untreated high-risk DLBCL it produced high complete response rates (ASH 2024). The phase 3 GOLSEEK-1 trial is enrolling patients with IPI 3 to 5 disease to compare golcadomide plus R-CHOP with R-CHOP alone, with readout expected in 2027 to 2028. BMS develops it. It is a next-generation lenalidomide-like pill designed to be added to standard first-line chemotherapy.","status":"phase-3","asOf":"2026-09-07","links":[{"label":"ClinicalTrials.gov NCT06356129 (GOLSEEK-1)","url":"https://clinicaltrials.gov/study/NCT06356129"}],"tags":[],"related":[],"cancers":["dlbcl"],"sections":[],"technologies":["protac-degrader"],"targets":["ikzf1","ikzf3"],"drugs":[],"companies":["bms"],"institutions":[],"pathways":[],"terms":[],"trials":["golseek-1","nct06911502","nct06090539","nct03930953","nct06425302"],"people":[],"bottlenecks":[],"keyPapers":[],"journals":[],"dependsOn":[],"notes":[],"code":"CC-99282, BMS-986369","modality":"Molecular glue degrader (CELMoD, IKZF1/3)","mechanism":"Binds cereblon and recruits IKZF1/IKZF3 for ubiquitination and proteasomal degradation; downregulates IRF4/MYC and stimulates T cells.","approvals":[],"mechanismSteps":[],"toxicity":[],"access":[],"regulatoryEvents":[]},"route":"/drugs/golcadomide/","neighbours":{"cancer":[{"id":"dlbcl","kind":"cancer","name":"Diffuse large B-cell lymphoma","route":"/cancers/dlbcl/"},{"id":"follicular-lymphoma","kind":"cancer","name":"Follicular lymphoma","route":"/cancers/follicular-lymphoma/"}],"technology":[{"id":"protac-degrader","kind":"technology","name":"PROTACs & molecular glues (targeted protein degradation)","route":"/technologies/protac-degrader/"}],"target":[{"id":"ikzf1","kind":"target","name":"IKZF1 (Ikaros)","route":"/targets/ikzf1/"},{"id":"ikzf3","kind":"target","name":"IKZF3 (Aiolos)","route":"/targets/ikzf3/"},{"id":"irf4","kind":"target","name":"IRF4","route":"/targets/irf4/"}],"company":[{"id":"bms","kind":"company","name":"Bristol Myers Squibb","route":"/companies/bms/"}],"trial":[{"id":"nct06835530","kind":"trial","name":"A Combination of Rituximab and CC-99282 as Front-line Therapy for Older Frail Patients With Diffuse Large B-cells Non-Hodgkin Lymphoma Evaluated With a Simplified Geriatric Assessment (sGA): a Phase II Study of the Fondazione Italiana Linfomi (FIL)","route":"/trials/nct06835530/"},{"id":"nct03930953","kind":"trial","name":"A Safety and Preliminary Efficacy Study of CC-99282, Alone and in Combination With Anti-lymphoma Agents in Participants With Relapsed or Refractory No","route":"/trials/nct03930953/"},{"id":"nct06090539","kind":"trial","name":"A Study to Assess BMS-986458 Alone and in Combination With Anti-lymphoma Agents in Relapsed/Refractory Non-Hodgkin Lymphomas","route":"/trials/nct06090539/"},{"id":"nct06911502","kind":"trial","name":"A Study to Compare the Efficacy and Safety of Golcadomide in Combination With Rituximab (Golca + R) vs Investigator's Choice in Participants With Rela","route":"/trials/nct06911502/"},{"id":"nct06425302","kind":"trial","name":"A Study to Evaluate the Efficacy and Safety of Golcadomide in Combination With Rituximab in Participants With Newly Diagnosed Advanced Stage Follicular Lymphoma","route":"/trials/nct06425302/"},{"id":"golseek-1","kind":"trial","name":"GOLSEEK-1","route":"/trials/golseek-1/"}],"pathway":[{"id":"ubiquitin-proteasome-system","kind":"pathway","name":"Ubiquitin-proteasome system & protein homeostasis","route":"/pathways/ubiquitin-proteasome-system/"}],"roadmap":[{"id":"epigenetics-roadmap","kind":"roadmap","name":"Epigenetic therapy roadmap: loosening silenced genes → mutation-specific enzymes → editing the epigenome","route":"/roadmaps/epigenetics-roadmap/"}]}}
