## Notes

- Pancreatic cohort of the phase 1/2 (NEJM 2026, Wolpin et al.): 168 previously treated patients with RAS-mutated pancreatic ductal adenocarcinoma received daraxonrasib at 300 mg or less; treatment-related adverse events of any grade in 96 percent and grade 3 or higher in 30 percent (rash, diarrhoea, nausea, stomatitis, vomiting, fatigue the commonest). In 26 patients with RAS G12 mutations treated second line at 300 mg the response rate was 35 percent with median progression-free survival 8.5 and overall survival 13.1 months; in 38 with G12, G13 or Q61 mutations 29 percent, 8.1 and 15.6 months. These figures set the 300 mg phase 3 dose for RASolute 302.
